New findings on personalized medicine for rare jaw tumors
Researchers at Charité – Universitätsmedizin Berlin have reported new findings on the personalized treatment of ameloblastoma. In a prospective observational study, they investigated whether targeted therapies can shrink these rare, benign jaw tumors before surgery, allowing for less extensive surgical treatment. Here, first author Dr. Dr. Elena Hofmann and study lead Dr. Max Schmidt answer questions about their research findings.
What was the research question or scientific inquiry behind your study?
Ameloblastoma is a rare, locally aggressive jaw tumor that mainly affects young adults and occasionally children. Standard treatment often requires extensive jaw surgery, resulting in functional and cosmetic impairment. We investigated whether comprehensive molecular tumor profiling could identify targeted therapies capable of shrinking tumors before surgery and thereby enable less extensive, function-preserving procedures.
How did you approach the topic?
We performed comprehensive molecular tumor profiling in 14 patients with ameloblastoma. The results were reviewed by an interdisciplinary Molecular Tumor Board to evaluate personalized treatment options. Eleven patients received matched targeted therapies. We then assessed how well the tumors responded to treatment and whether tumor shrinkage enabled less extensive surgery.
What did you discover?
In ten of the eleven patients treated with targeted therapies, the tumors shrank markedly. In the four patients who have undergone surgery so far, the procedures were considerably less extensive than originally planned. We were able to preserve larger portions of jawbone and teeth, resulting in better functional and aesthetic outcomes. Our findings highlight the potential of precision medicine for patients with rare tumors.
What’s your takeaway?
Our findings support the use of molecular tumor profiling at the time of diagnosis for patients with ameloblastoma. Targeted therapies may shrink tumors before surgery, allowing for less invasive procedures while preserving jaw function and facial appearance. These promising results now need to be confirmed in prospective interventional clinical trials.
